I am very satisfied with the constructions (double mutants) made by Vectorbuilder. The service is very serious and rigorous. They started by verifying our plasmid by doing digestions. As this was not enough, they even sequenced part of the plasmid, which then ensured the success of the constructs. Constant email exchanges for verification steps were ensured with almost instantaneous responsiveness! Quite impressive! Once the sequence was clarified, production was not long, and we were able to do our experiments under excellent conditions. The results were as good as we expected. VectorBuilder's AAV serotype panel helped us to choose the best expressing serotype for our brain area and animal model of interest. By piloting injections using a selection of serotypes provided in the panel, we obtained invaluable pilot data before ordering a large scale virus production. This allowed us to save a considerable amount of time and resources. The technical support was great, they answer our questions promptly provided great advice. VectorBuilder technical support team further help us designing and cloning our AAV viral vector. Our preliminary data allowed to map retrogradely labelled neurons in the amygdala using light-sheet microscopy. We would not hesitate to use VectorBuilder for our next projects

Two years ago, when another CRO was struggling to provide the requested clones that I ordered, I reached out to Vector Builder to try to make them for me. We applied
CRISPR cas9 nickase in our study. VectorBuilder helped us design the gRNAs and construct plasmids with double gRNAs and double selection markers (EGFP/puromycin). The gRNA specificity is quite good, better than available online design tools. We got the knockout cell lines in the first run of electroporation with no off-target events. I would recommend VectorBuilder to CRISPR cas9 users.Jie Qin

We ordered adenovirus from VectorBuilder to overexpress our protein of interest in primary mouse hepatocytes. The ordering process, including vector design, was very fast and easy. We received the plasmid and adenovirus within the time frame that was stated during the ordering process and the transduction efficiency of the adenovirus was very high, enabling us to use a low MOI. As a bonus, we also received the control Ad-EGFP adenovirus for free. In the future, I will not use any more siRNA or classical transfection methods on primary mouse hepatocytes, just adenoviruses from VB! I can highly recommend VectorBuilder's services for vector design and adenovirus production.
